Cryo-electron microscopy (cryo-EM) is a powerful tool for studying biological structures at molecular level, but measurement quality strongly depends on sample preparation. Preparing cryo-EM grids is traditionally a delicate, operator-dependent process, making consistent results difficult to achieve.
Together with CryoSol, Demcon developed the VitroJet: an automated sample preparation system that integrates grid treatment, sample deposition, environmental control, inspection and vitrification into one controlled workflow. This improves reproducibility while reducing operator dependency and the use of valuable biological samples.
From delicate procedure to reproducible workflow
Cryo-EM sample preparation requires several critical steps to be performed accurately and in rapid succession. Extremely small sample volumes must be deposited onto fragile grids, temperature and humidity must remain stable, and the sample must be vitrified rapidly to preserve the biological structure.
The engineering challenge was to automate this process without compromising precision or stability. This required:
- Precise handling and positioning of fragile cryo-EM grids
- Accurate deposition of very small sample volumes
- Controlled temperature and humidity
- Fast and synchronized processing and vitrification
- Inline inspection and process verification
integrating the complete preparation process.
The VitroJet combines these functions in one automated platform. Automated grid handling is combined with pin-printing technology to deposit extremely thin liquid layers using very small sample volumes. Embedded plasma treatment supports consistent grid wettability, while optical inspection and imaging enable process verification. Environmental control, workflow automation and rapid vitrification complete the process.
Bringing these technologies together reduces manual handling and process variability. For researchers, this means more reproducible samples, more efficient use of scarce biological material and less dependency on individual operator experience.
engineering a reproducible sample preparation workflow.
Demcon developed and integrated the key technologies required for the automated workflow, including precision mechatronics, environmental and thermal control, optical inspection, automation software, cryogenic technology and overall system architecture.
The VitroJet shows how system integration can turn a complex, operator-dependent laboratory procedure into a robust and repeatable workflow. The result is more consistent sample preparation and a more efficient process, allowing researchers to focus on their research rather than the preparation itself.
